    Description In the field of “Redox biology in the cardiovascular system” the research group of Professor Andreas Daiber offers the following PhD project:
    Redox regulation of epigenetic pathways and oxidative DNA damage in the cardiovascular system

    PhD project proposal: With the present proposal we suggest to use animal models with reduced or increased ROS formation (e.g. p47phox-/- and Cu,Zn-SOD-/- mice) along with clinically relevant oxidative stress challenges (e.g. pharmacologically- or genetically-induced hypertension, diabetes, obesity) in order to identify novel “redox switches” in epigenetic pathways [Mikhed et al. Redox Biology 2015] and oxidative damage of DNA [Mikhed et al. IJMS 2015; Mikhed et al. Basic Res. Cardiol. 2016]. Relevant endpoints will include oxidized DNA lesions (8-oxoguanine by immunostaining), strand breaks (by comet assay) and histone marks (e.g. H3K4me and H3K9ac). The cardiovascular phenotype will be characterized by blood pressure, vascular function, markers of inflammation and oxidative stress parameters.
    In addition, we will assess redox modifications of enzymes involved in epigenetic pathways and DNA repair (e.g. dityrosine, 3-nitrotyrosine, S-glutathionylation, S-nitrosation) and determine their functional consequences for gene expression. In conclusion, the present study aims at the identification of the regulatory and damaging role of reactive oxygen and nitrogen species in epigenetic pathways and genome stability for potential translation of these findings into clinical use for the prevention of cardiovascular risk factors and treatment of cardiovascular diseases [Mikhed et al. Redox Biology 2015; Daiber et al. Br. J. Pharmacol. 2016].
    The successful applicant should have a sound background in (bio)chemistry.
